Abstract

The flow cytometry has been used worldwide as a prognostic factor for breast câncer.
However, the feasibility ofthe method using aspirates obtainedby fine needie aspiration cytoiogy
(FNAC) remains controverse. Objective: Comparing the aspirates generated by FNAC with
those from the paraffin embedded specimen, through fiow cytometry. Patients and methods:
The study was performed in 44 fresh surgicai specimens containing breast tumors, with the
taps using both methods: autovacuum device and sirynge pistoi hoider. Each specimen was
punched twice with a fine needie. At a iater date, these specimens were fixated andpreserved
in paraffin. Both the aspirates and the histoiogy material, without the paraffin coating, were
prepared for fiow cytometry. A BD FACScan, capab/e of reading 10,000 ce/ls per test was used.
Resu/ts: The mean for CV from aspirates was 6.05 (± 1.6) and for the surgicai specimen was
6.96 (± 2.13) (p < 0.05). The mean for DNA index was 1.6 (± 0.5) from aspirates and 1.49
(± 0.42) of the surgicai specimens (NS). The mean for aspirates' S-phase was 7.2 (± 6.34)
and 5.85 (± 4.73) for the specimen (NS). For aneuploidy, the mean was 38.2 (± 24.7) and
29.8 (± 20) for aspirates and specimen, respectivei/y (NS). Condusion: The aspirates generated
from FNAC are very reliable for fiow cytometry study and they might be betterthan the paraffin
embedded tissue.
